Više od milijardu ljudi diljem svijeta pati od migrene. Točan joj se uzrok ne zna, ali se vjeruje da je u osnovi migrene, patofiziološki, neurovaskularni poremećaj koji se odnosi na mehanizme unutar mozga i vaskularnih struktura.

Najveća studija dosad uključivala je vodeće grupe za istraživanje migrene u Europi, Australiji i Sjedinjenim Državama koje su zajedno radile na prikupljanju genetskih podataka od više od 873.000 sudionika studije, od kojih je 102.000 imalo migrenu.

Otkrića su objavljena u časopisu Nature Genetics, i pokazala su daleko širu genetsku arhitekturu za podtipove migrene nego što je ranije bilo poznato, i čak je 120 regija genoma identificirano u vezi s rizikom od migrene, piše Portal Zdravlje.

Prethodna istraživanja su pokazala da genetski čimbenici značajno pridonose riziku od migrene. Međutim, dugo se raspravljalo o tome imaju li dvije glavne vrste migrene – migrena s aurom i migrena bez aure – sličnu genetsku pozadinu.

Istraživači iz Međunarodnog konzorcija za genetiku glavobolje sakupili su veliki skup podataka kako bi proveli studiju povezanosti na razini genoma, tražeći genetske varijante koje su bile češće kod onih koji su imali migrenu općenito, ili jedan od dva glavna tipa migrene.

Rezultati su pokazali da podtipovi migrene imaju i zajedničke čimbenike rizika i one koji se čine specifičnim za jedan podtip. Analize su istaknule tri varijante rizika koje se čine specifične za migrenu s aurom i dvije koje se čine specifične za migrenu bez aure.

Neuronski i vaskularni genetski čimbenici

“Osim desetaka novih regija genoma za preciznija istraživanja, studija pruža prvu smislenu mogućnost za procjenu zajedničkih i različitih genetskih komponenti u dva glavna podtipa migrene”, rekla je prva autorica studije, Heidi Hautakangas iz Instituta za molekularnu medicinu Finska, Sveučilište u Helsinkiju.

Rezultati su podržali koncept prema kojem migrenu uzrokuju i neuronski i vaskularni genetski čimbenici, jačajući stav da je migrena uistinu neurovaskularni poremećaj.

Važan je potencijal istraživanja da ukaže na nove terapije protiv migrene.

Budući da je migrena u svijetu drugi najveći faktor koji doprinosi godinama života s invaliditetom, očito postoji velika potreba za novim tretmanima. Posebno zanimljiv nalaz bio je identifikacija genomskih rizičnih regija što sadrže gene koji kodiraju mete za nedavno razvijene terapeutike specifične za migrenu.

Jedna od novoidentificiranih regija sadrži gene koji kodiraju peptid povezan s genom kalcitonina, molekulu uključenu u napade migrene i blokiranu nedavno uvedenim lijekovima za inhibitore migrene CGRP. Još jedna rizična regija pokriva gen koji kodira receptor za serotonin 1F, također cilj za nove lijekove specifične za migrenu, piše Portal Zdravlje.

Više od milijardu ljudi diljem svijeta pati od migrene. Točan joj se uzrok ne zna, ali se vjeruje da je u osnovi migrene, patofiziološki, neurovaskularni poremećaj koji se odnosi na mehanizme unutar mozga i vaskularnih struktura.

Najveća studija dosad uključivala je vodeće grupe za istraživanje migrene u Europi, Australiji i Sjedinjenim Državama koje su zajedno radile na prikupljanju genetskih podataka od više od 873.000 sudionika studije, od kojih je 102.000 imalo migrenu.

Otkrića su objavljena u časopisu Nature Genetics, i pokazala su daleko širu genetsku arhitekturu za podtipove migrene nego što je ranije bilo poznato, i čak je 120 regija genoma identificirano u vezi s rizikom od migrene, piše Portal Zdravlje.

Prethodna istraživanja su pokazala da genetski čimbenici značajno pridonose riziku od migrene. Međutim, dugo se raspravljalo o tome imaju li dvije glavne vrste migrene – migrena s aurom i migrena bez aure – sličnu genetsku pozadinu.

Istraživači iz Međunarodnog konzorcija za genetiku glavobolje sakupili su veliki skup podataka kako bi proveli studiju povezanosti na razini genoma, tražeći genetske varijante koje su bile češće kod onih koji su imali migrenu općenito, ili jedan od dva glavna tipa migrene.

Rezultati su pokazali da podtipovi migrene imaju i zajedničke čimbenike rizika i one koji se čine specifičnim za jedan podtip. Analize su istaknule tri varijante rizika koje se čine specifične za migrenu s aurom i dvije koje se čine specifične za migrenu bez aure.

Neuronski i vaskularni genetski čimbenici

“Osim desetaka novih regija genoma za preciznija istraživanja, studija pruža prvu smislenu mogućnost za procjenu zajedničkih i različitih genetskih komponenti u dva glavna podtipa migrene”, rekla je prva autorica studije, Heidi Hautakangas iz Instituta za molekularnu medicinu Finska, Sveučilište u Helsinkiju.

Rezultati su podržali koncept prema kojem migrenu uzrokuju i neuronski i vaskularni genetski čimbenici, jačajući stav da je migrena uistinu neurovaskularni poremećaj.

Važan je potencijal istraživanja da ukaže na nove terapije protiv migrene.

Budući da je migrena u svijetu drugi najveći faktor koji doprinosi godinama života s invaliditetom, očito postoji velika potreba za novim tretmanima. Posebno zanimljiv nalaz bio je identifikacija genomskih rizičnih regija što sadrže gene koji kodiraju mete za nedavno razvijene terapeutike specifične za migrenu.

Jedna od novoidentificiranih regija sadrži gene koji kodiraju peptid povezan s genom kalcitonina, molekulu uključenu u napade migrene i blokiranu nedavno uvedenim lijekovima za inhibitore migrene CGRP. Još jedna rizična regija pokriva gen koji kodira receptor za serotonin 1F, također cilj za nove lijekove specifične za migrenu, piše Portal Zdravlje.
