Unatoč stoljećima istraživanja i napretka u medicini, još uvijek postoje mnoge misterije koje ostaju neriješene, a glavna među njima je razumijevanje uzroka starenja i kako ih možemo usporiti ili preokrenuti. No, nova studija tima znanstvenika iz Njemačke, objavljena u znanstvenom časopisu Nature, možda je konačno pronašla odgovore na ova pitanja.
Istraživači sa Sveučilišta u Kölnu ne samo da su otkrili da transkripcija gena - proces u kojem stanica stvara RNA kopiju lanca DNK - postaje brža s godinama, nego da je i manje precizna i sklonija pogreškama. Također su otkrili da nam određeni procesi mogu pomoći da preokrenemo taj postupak.
„Ovo je do sada jedini trenutak euforije u mom životu. Mislim, ovo je vrsta otkrića do kojeg ne dolazite svaki dan", rekao je dr. Andreas Beyer, vodeći istraživač, nazivajući rezultate "velikim otkrićem".

Prije nego što je Beyer sa svojim timom započeo svoj istraživački projekt prije 10 godina, tipična istraživanja starenja samo su "promatrala diferencijalnu ekspresiju gena", pojasnio je. Prethodna istraživanja, objašnjava on, postavljala su pitanja poput: "Koji se geni uključuju, a koji isključuju tijekom starenja?" i "Kako se to mijenja regulacija ili metabolizam u stanici?".

Ali nitko nije pitao kako se sam proces transkripcije mijenja kako starimo, i linija istraživanja koja bi mogla pružiti uvide koji nam u konačnici mogu pomoći da preokrenemo ili zaustavimo propadanje. Transkripcija je temeljna za Beyerovo istraživanje budući je proces u kojem stanica stvara RNA kopiju dijela DNA. Ova kopija je važna jer nosi genetske informacije potrebne za stvaranje novih proteina u stanicama. Proteini određuju zdravlje i funkciju stanica, a stanice pak čine strukturu svih živih bića.
„Tijekom našeg života, naše se stanice regeneriraju, ali svaka stanica je različita, a ono što ih čini različitima su različiti geni koji su u njoj aktivirani, objašnjava Beyer i dodaje kako se „ova aktivacija naziva transkripcija".
Budući da geni određuju svrhu stanica, njihova transkripcija mora biti besprijekorna.

"Potrebno je stvoriti točnu količinu transkripata za svaki gen i imati točnu kopiju sekvence gena, ali također morate aktivirati točne gene koje stanica treba kako bi ispravno funkcionirala", rekao je Beyer.
U ljudskom tijelu postoji mnogo različitih vrsta stanica: živčane, mišićne, krvne, stanice kože i tako dalje. Budući da svaka stanica obavlja različitu funkciju, u svakom tipu stanice aktivira se (transkribira) drugi skup gena.
"Stroj" - kako ga naziva Beyer - odgovoran za izradu transkripcijske kopije genetskog slijeda naziva se Pol II (RNA polimeraza II). Ono što je njegov tim otkrio jest da se proces transkripcije ubrzava kako starimo, a ovaj ubrzani proces uzrokuje da Pol II čini više pogrešaka, što dovodi do suštinski loših kopija koje mogu dovesti do brojnih bolesti.
"Ako Pol II postane prebrz, čini više pogrešaka i tada sekvenca više nije identična sekvenci genoma. Posljedice su slične onima koje imate kada postoje mutacije u samom genomu”, rekao je Beyer.
I restrikcija kalorija pomaže
Prethodna istraživanja već su dokazala da dijete s niskim unosom kalorija i inhibiranje inzulinske signalizacije - blokiranje signala između inzulina i stanica - mogu usporiti starenje i produljiti životni vijek mnogih životinja.
U svojim eksperimentima, Beyerov tim je istraživao imaju li ove metode utjecaj na usporavanje brzine Pol II i smanjenje broja pogrešnih kopija. Istraživanje - zajednička suradnja 26 znanstvenika u šest različitih laboratorija - prvo se provodilo s crvima, miševima i vinskim mušicama genetski modificiranim kako bi se inhibirala inzulinska signalizacija, kao i s miševima na niskokaloričnoj dijeti kako bi se odredila učinkovitost transkripcije stanica u starijoj dobi. U oba slučaja, Pol II je reagirao i putovao sporije, čineći manje grešaka.

Beyer i njegov tim zatim su pratili preživljavanje mušica i crva koji su nosili mutaciju koja je usporavala Pol II, i životinje su živjele 10 do 20 posto duže u odnosu na one bez mutacije. Kada su istraživači upotrijebili uređivanje gena da bi opozvali mutacije kod crva - životni vijek životinja skratio se, uspostavljajući uzročnu vezu.
Kako bi testirali svoj eksperiment na ljudima, radili su s uzorcima krvi mladih i starih osoba.
"I kada smo usporedili mlade stanice s vrlo starim stanicama, in vitro, dobili smo potpuno iste rezultate", rekao je Argyris Papantonis, jedan od istraživača, za Euronews Next.
Rezultati među različitim vrstama potvrđuju da je to „zaista opći fenomen koji se odnosi na starenje, a ne samo specifičan za pojedinačni model, primjerice muhe", istaknuo je Beyer.
"Naša studija kaže da bi, na primjer, zdrava prehrana ili ova intervencija ograničenja kalorija poboljšala kvalitetu transkripcije proizvodnje RNK u stanici. A to bi onda dugoročno imalo korisne učinke za stanice. Nalazi bi mogli pomoći u sprječavanju manifestacije raka, napominje Papantonis, jer je "to bolest kasne životne dobi zbog pogrešaka. Ograničavanje pogrešaka može biti način ograničavanja pojave raka ili bolesti kasne životne dobi".
„Oni nam također mogu omogućiti bolje razumijevanje onoga što se događa kada starimo, i u konačnici bolje razumijevanje intervencija, što mislim da otvara nove mogućnosti za odgađanje starenja ili produženje zdravog starenja", zaključio je Beyer.

Unatoč stoljećima istraživanja i napretka u medicini, još uvijek postoje mnoge misterije koje ostaju neriješene, a glavna među njima je razumijevanje uzroka starenja i kako ih možemo usporiti ili preokrenuti. No, nova studija tima znanstvenika iz Njemačke, objavljena u znanstvenom časopisu Nature, možda je konačno pronašla odgovore na ova pitanja.
Istraživači sa Sveučilišta u Kölnu ne samo da su otkrili da transkripcija gena - proces u kojem stanica stvara RNA kopiju lanca DNK - postaje brža s godinama, nego da je i manje precizna i sklonija pogreškama. Također su otkrili da nam određeni procesi mogu pomoći da preokrenemo taj postupak.
„Ovo je do sada jedini trenutak euforije u mom životu. Mislim, ovo je vrsta otkrića do kojeg ne dolazite svaki dan", rekao je dr. Andreas Beyer, vodeći istraživač, nazivajući rezultate "velikim otkrićem".
"Na Twitteru se diže prava oluja. Neki kolege su vrlo uzbuđeni", rekao je Beyer za Euronews Next.
Prije nego što je Beyer sa svojim timom započeo svoj istraživački projekt prije 10 godina, tipična istraživanja starenja samo su "promatrala diferencijalnu ekspresiju gena", pojasnio je. Prethodna istraživanja, objašnjava on, postavljala su pitanja poput: "Koji se geni uključuju, a koji isključuju tijekom starenja?" i "Kako se to mijenja regulacija ili metabolizam u stanici?".

Ali nitko nije pitao kako se sam proces transkripcije mijenja kako starimo, i linija istraživanja koja bi mogla pružiti uvide koji nam u konačnici mogu pomoći da preokrenemo ili zaustavimo propadanje. Transkripcija je temeljna za Beyerovo istraživanje budući je proces u kojem stanica stvara RNA kopiju dijela DNA. Ova kopija je važna jer nosi genetske informacije potrebne za stvaranje novih proteina u stanicama. Proteini određuju zdravlje i funkciju stanica, a stanice pak čine strukturu svih živih bića.
„Tijekom našeg života, naše se stanice regeneriraju, ali svaka stanica je različita, a ono što ih čini različitima su različiti geni koji su u njoj aktivirani, objašnjava Beyer i dodaje kako se „ova aktivacija naziva transkripcija".
Budući da geni određuju svrhu stanica, njihova transkripcija mora biti besprijekorna.

"Potrebno je stvoriti točnu količinu transkripata za svaki gen i imati točnu kopiju sekvence gena, ali također morate aktivirati točne gene koje stanica treba kako bi ispravno funkcionirala", rekao je Beyer.
U ljudskom tijelu postoji mnogo različitih vrsta stanica: živčane, mišićne, krvne, stanice kože i tako dalje. Budući da svaka stanica obavlja različitu funkciju, u svakom tipu stanice aktivira se (transkribira) drugi skup gena.
"Stroj" - kako ga naziva Beyer - odgovoran za izradu transkripcijske kopije genetskog slijeda naziva se Pol II (RNA polimeraza II). Ono što je njegov tim otkrio jest da se proces transkripcije ubrzava kako starimo, a ovaj ubrzani proces uzrokuje da Pol II čini više pogrešaka, što dovodi do suštinski loših kopija koje mogu dovesti do brojnih bolesti.
"Ako Pol II postane prebrz, čini više pogrešaka i tada sekvenca više nije identična sekvenci genoma. Posljedice su slične onima koje imate kada postoje mutacije u samom genomu”, rekao je Beyer.
I restrikcija kalorija pomaže
Prethodna istraživanja već su dokazala da dijete s niskim unosom kalorija i inhibiranje inzulinske signalizacije - blokiranje signala između inzulina i stanica - mogu usporiti starenje i produljiti životni vijek mnogih životinja.
U svojim eksperimentima, Beyerov tim je istraživao imaju li ove metode utjecaj na usporavanje brzine Pol II i smanjenje broja pogrešnih kopija. Istraživanje - zajednička suradnja 26 znanstvenika u šest različitih laboratorija - prvo se provodilo s crvima, miševima i vinskim mušicama genetski modificiranim kako bi se inhibirala inzulinska signalizacija, kao i s miševima na niskokaloričnoj dijeti kako bi se odredila učinkovitost transkripcije stanica u starijoj dobi. U oba slučaja, Pol II je reagirao i putovao sporije, čineći manje grešaka.

Beyer i njegov tim zatim su pratili preživljavanje mušica i crva koji su nosili mutaciju koja je usporavala Pol II, i životinje su živjele 10 do 20 posto duže u odnosu na one bez mutacije. Kada su istraživači upotrijebili uređivanje gena da bi opozvali mutacije kod crva - životni vijek životinja skratio se, uspostavljajući uzročnu vezu.
Kako bi testirali svoj eksperiment na ljudima, radili su s uzorcima krvi mladih i starih osoba.
"I kada smo usporedili mlade stanice s vrlo starim stanicama, in vitro, dobili smo potpuno iste rezultate", rekao je Argyris Papantonis, jedan od istraživača, za Euronews Next.
Rezultati među različitim vrstama potvrđuju da je to „zaista opći fenomen koji se odnosi na starenje, a ne samo specifičan za pojedinačni model, primjerice muhe", istaknuo je Beyer.
"Naša studija kaže da bi, na primjer, zdrava prehrana ili ova intervencija ograničenja kalorija poboljšala kvalitetu transkripcije proizvodnje RNK u stanici. A to bi onda dugoročno imalo korisne učinke za stanice. Nalazi bi mogli pomoći u sprječavanju manifestacije raka, napominje Papantonis, jer je "to bolest kasne životne dobi zbog pogrešaka. Ograničavanje pogrešaka može biti način ograničavanja pojave raka ili bolesti kasne životne dobi".
„Oni nam također mogu omogućiti bolje razumijevanje onoga što se događa kada starimo, i u konačnici bolje razumijevanje intervencija, što mislim da otvara nove mogućnosti za odgađanje starenja ili produženje zdravog starenja", zaključio je Beyer.
