Novo eksperimentalno cjepivo protiv raka pokazalo je ohrabrujuće rezultate u kliničkoj studiji na malom broju pacijenata s tumorima gušterače i debelog crijeva, priopćili su znanstvenici na ovogodišnjem kongresu Američkog društva za kliničku onkologiju (ASCO) u Chicagu.

Cjepivo ELI-002 razvila je mala biotehnološka kompanija Elicio Therapeutics iz Bostona, a namijenjeno je pacijentima čiji je rak uzrokovan mutacijom gena KRAS, odgovornom za 30 do 50 posto slučajeva kolorektalnih tumora.

- Primarni cilj ovoga kliničkog pokusa bio je pokazati sigurnost i izvodljivost - rekla je za DailyMail dr. Eileen O‘Reilly, gastrointestinalna onkologinja s Memorial Sloan Kettering Cancer Center u New Yorku koja je vodila kliničku studiju. U studiju su bila uključena 25 pacijenata kojima je dijagnosticiran rak gušterače ili debelog crijeva. Svima su kirurški odstranili tumore i nisu pokazivali znakove maligne bolesti na snimkama. Međutim, još uvijek su imali tumorske biomarkere u krvi što znači da je rak i dalje prisutan što dovodi do gotovo neizbježnog rizika od povratka bolesti.

Pacijenti su zatim dobili cjepivo ELI-002: ovo cjepivo koristi dva peptida, G12D i G12R. Oni se vežu za albumin, protein u tijelu koji prenosi masne kiseline do limfnih čvorova. Albumin pomaže cjepivu da putuje izravno do limfnih čvorova, aktivirajući imunološki sustav tako da se stvaraju bijele krvne stanice, koje pomažu u borbi protiv tumorskih markera u krvi pacijenata.

Cjepivo se ubrizgava u obje ruke i obje noge, ukupno četiri injekcije svaki tjedan tijekom četiri tjedna. Pacijenti su nastavili s ovim procesom svaki drugi tjedan tijekom dva tjedna, a zatim su pauzirali prije početka tromjesečnog docjepljivanja. Faza docjepljivanja je neophodna jer nakon nekog vremena imunološki odgovor na cjepivo slabi. Kada su dobili najviše doze cjepiva, svi pacijenti su imali imunološki odgovor. To sugerira da bi cjepivo moglo pomoći u smanjenju tumorskih markera kod pacijenata s dugotrajnom bolešću.

Cjepivo ELI-2 nije personalizirano, tj. nije prilagođeno svakom pojedinom pacijentu.

- Postoje ogromne praktične prednosti ovih vrsta pristupa u odnosu na personalizirana cjepiva protiv raka - rekla je dr. O‘Reilly istaknuvši da su u tijeku pripreme za drugu fazu kliničke studije.

Dobri rezultati prve faze kliničkog ispitivanja pružaju veliku nadu jer je rak gušterače jedan od najsmrtonosnijih: u prosjeku, samo 12,5 posto preživi pet godina nakon dijagnoze. Nadalje, broj slučajeva raka debelog crijeva u nizu zemalja je u porastu te se sve češće dijagnosticira u mlađoj dobi te u poodmaklom stadiju bolesti.

Novo eksperimentalno cjepivo protiv raka pokazalo je ohrabrujuće rezultate u kliničkoj studiji na malom broju pacijenata s tumorima gušterače i debelog crijeva, priopćili su znanstvenici na ovogodišnjem kongresu Američkog društva za kliničku onkologiju (ASCO) u Chicagu.

Cjepivo ELI-002 razvila je mala biotehnološka kompanija Elicio Therapeutics iz Bostona, a namijenjeno je pacijentima čiji je rak uzrokovan mutacijom gena KRAS, odgovornom za 30 do 50 posto slučajeva kolorektalnih tumora.

- Primarni cilj ovoga kliničkog pokusa bio je pokazati sigurnost i izvodljivost - rekla je za DailyMail dr. Eileen O‘Reilly, gastrointestinalna onkologinja s Memorial Sloan Kettering Cancer Center u New Yorku koja je vodila kliničku studiju. U studiju su bila uključena 25 pacijenata kojima je dijagnosticiran rak gušterače ili debelog crijeva. Svima su kirurški odstranili tumore i nisu pokazivali znakove maligne bolesti na snimkama. Međutim, još uvijek su imali tumorske biomarkere u krvi što znači da je rak i dalje prisutan što dovodi do gotovo neizbježnog rizika od povratka bolesti.

Pacijenti su zatim dobili cjepivo ELI-002: ovo cjepivo koristi dva peptida, G12D i G12R. Oni se vežu za albumin, protein u tijelu koji prenosi masne kiseline do limfnih čvorova. Albumin pomaže cjepivu da putuje izravno do limfnih čvorova, aktivirajući imunološki sustav tako da se stvaraju bijele krvne stanice, koje pomažu u borbi protiv tumorskih markera u krvi pacijenata.

Cjepivo se ubrizgava u obje ruke i obje noge, ukupno četiri injekcije svaki tjedan tijekom četiri tjedna. Pacijenti su nastavili s ovim procesom svaki drugi tjedan tijekom dva tjedna, a zatim su pauzirali prije početka tromjesečnog docjepljivanja. Faza docjepljivanja je neophodna jer nakon nekog vremena imunološki odgovor na cjepivo slabi. Kada su dobili najviše doze cjepiva, svi pacijenti su imali imunološki odgovor. To sugerira da bi cjepivo moglo pomoći u smanjenju tumorskih markera kod pacijenata s dugotrajnom bolešću.

Cjepivo ELI-2 nije personalizirano, tj. nije prilagođeno svakom pojedinom pacijentu.

- Postoje ogromne praktične prednosti ovih vrsta pristupa u odnosu na personalizirana cjepiva protiv raka - rekla je dr. O‘Reilly istaknuvši da su u tijeku pripreme za drugu fazu kliničke studije.

Dobri rezultati prve faze kliničkog ispitivanja pružaju veliku nadu jer je rak gušterače jedan od najsmrtonosnijih: u prosjeku, samo 12,5 posto preživi pet godina nakon dijagnoze. Nadalje, broj slučajeva raka debelog crijeva u nizu zemalja je u porastu te se sve češće dijagnosticira u mlađoj dobi te u poodmaklom stadiju bolesti.
