Francuz već više od 40 godina boluje od genetski uvjetovanog očnog stanja, retinitis pigmentosa, a koje uzrokuje umiranje fotoosjetljivih stanica, piše Jutarnji list.

Znanstvenici su došli do otkrića koje bi moglo olakšati život milijuna ljudi diljem svijeta - potpuno slijepom muškarcu vid je djelomično vraćen nakon što je podvrgnut terapiji proteinom koji je prvotno otkriven kod algi. Biološka terapija, takozvana optogenetika, poseban je proces pri kojem se proteini koriste kako bi se organizmu vratila kontrola nad stanicama u pozadini oka. 

Kako piše BBC citirajući časopis Nature Medicine, potpuno slijep muškarac pomalo je počeo raspoznavati oblike, poput obojanih linija na pješačkom prijelazu, "zebri". Da je tretman uspio postalo je jasno kada je muškarac iz Francuske, čiji identitet nije otkriven, mogao samostalno raspoznavati, brojati i uzimati predmete sa stola. 

Francuz već više od 40 godina boluje od genetski uvjetovanog očnog stanja koje zajednički nazivamo retinitis pigmentosa, a koje uzrokuje umiranje fotoosjetljivih stanica na površini retine, odnosno, mrežnice. S tim se problemom diljem svijeta suočava više od dva milijuna ljudi, a iako je potpuna sljepoća vrlo rijetka, muškarac koji je podvrgnut eksperimentalnom liječenju prije dva desetljeća posve je izgubio vid. 

Optogenika

BBC navodi kako je optogenetika razmjerno nova u medicini, ali se dugo razmatra na području neuroznanosti. Koristi svjetlo kako bi precizno utjecalo na aktivnosti stanica u mozgu, te je u ovom slučaju pomoglo da se jedno pacijentovo oko ponovno osposobi za prepoznavanje svjetla. Tehnika se, piše isti izvor, bazira na proteinima koje proizvode alge, nazvane channelrhodopsins, a koji mijenjaju način na koji se ponašaju kao odgovor na svjetlo. Mikrobi ih koriste kako bi približavali svjetlu.

Prvi korak u liječenju bila je genska terapija. Genetske instrukcije za proizvodnju rodopsina uzete su iz alga te su "predate" na korištenje stanicama u dubini mrežnice. Kada su pogođene snopom svjetla počele su slati električne signale mozgu. Postupak uspijeva samo kada se stanice izlože takozvanom jantarnom svjetlu, pa je pacijent nosio naočale s videokamerom na čelu i projektorom, kako bi se u pozadinu njegova oka mogla projicirati verzija onog što se događa u stvarnom svijetu. 

Bili su potrebni, ističu stručnjaci, mjeseci ustrajnog rada dok se u oku nisu stvorile dostatne količine rodopsina. Prvi znak da će terapija uspjeti bio je kada je pacijent prilikom šetnje odjednom počeo raspoznavati oblike na pješačkom prijelazu. 

Dr. José-Alain Sahel s pariškog Instituta za vid rekao je kako je pacijent isprva bio pomalo frustriran zbog činjenice da postupak traje toliko dugo. 

- Ali, kao što možete zamisliti, bio je vrlo uzbuđen kad je spontano počeo raspoznavati svari. Svi smo bili uzbuđeni. Još uvijek nema sjajan vid, no razlika između nepostojećeg i ograničenog vida nešto je što mijenja život - kazao je liječnik. 

Goleme mogućnosti

Botond Roska, sa sveučilišta u švicarskom Baselu rekao je kako nova saznanja dokazuju da je optogenetika terapija budućnosti kada je povrat vida u pitanju. 

Ipak, to je samo jedna od brojnih tehnika koje liječnici diljem svijeta isprobavaju kako bi osobama vratili vid, piše BBC. Mnoge se također bave popravkom genetskih problema koje uzrokuju probleme s vidom, no s retinitis pigmentosom to je posebno teško jer ju može uzrokovati mutacija više od 71 različitog gena. Neki znanstvenici pokušavaju oboljelima vratiti vid tako što povezuju videokameru s elektrodama implantiranim u pozadinu oka. 

Vjeruje se da su mogućnosti primjene optogenetike goleme, a neki zagovaraju upotrebu tehnike pri liječenju Parkinsonove bolesti, pa čak i kod nadvladavanja problema uzrokovanih moždanim udarom. 

Britanski profesor i stručnjak za bolesti mrežnice James Bainbridge, ustvrdio je kako je studija izuzetno obećavajuća, no da tek treba vidjeti rezultate na drugim ispitanicima, jer je sada provedena samo na jednom pacijentu.

Francuz već više od 40 godina boluje od genetski uvjetovanog očnog stanja, retinitis pigmentosa, a koje uzrokuje umiranje fotoosjetljivih stanica, piše Jutarnji list.

Znanstvenici su došli do otkrića koje bi moglo olakšati život milijuna ljudi diljem svijeta - potpuno slijepom muškarcu vid je djelomično vraćen nakon što je podvrgnut terapiji proteinom koji je prvotno otkriven kod algi. Biološka terapija, takozvana optogenetika, poseban je proces pri kojem se proteini koriste kako bi se organizmu vratila kontrola nad stanicama u pozadini oka. 

Kako piše BBC citirajući časopis Nature Medicine, potpuno slijep muškarac pomalo je počeo raspoznavati oblike, poput obojanih linija na pješačkom prijelazu, "zebri". Da je tretman uspio postalo je jasno kada je muškarac iz Francuske, čiji identitet nije otkriven, mogao samostalno raspoznavati, brojati i uzimati predmete sa stola.

S tim se problemom diljem svijeta suočava više od dva milijuna ljudi, a iako je potpuna sljepoća vrlo rijetka, muškarac koji je podvrgnut eksperimentalnom liječenju prije dva desetljeća posve je izgubio vid. 

Optogenika

BBC navodi kako je optogenetika razmjerno nova u medicini, ali se dugo razmatra na području neuroznanosti. Koristi svjetlo kako bi precizno utjecalo na aktivnosti stanica u mozgu, te je u ovom slučaju pomoglo da se jedno pacijentovo oko ponovno osposobi za prepoznavanje svjetla. Tehnika se, piše isti izvor, bazira na proteinima koje proizvode alge, nazvane channelrhodopsins, a koji mijenjaju način na koji se ponašaju kao odgovor na svjetlo. Mikrobi ih koriste kako bi približavali svjetlu.

Prvi korak u liječenju bila je genska terapija. Genetske instrukcije za proizvodnju rodopsina uzete su iz alga te su "predate" na korištenje stanicama u dubini mrežnice. Kada su pogođene snopom svjetla počele su slati električne signale mozgu. Postupak uspijeva samo kada se stanice izlože takozvanom jantarnom svjetlu, pa je pacijent nosio naočale s videokamerom na čelu i projektorom, kako bi se u pozadinu njegova oka mogla projicirati verzija onog što se događa u stvarnom svijetu. 

Bili su potrebni, ističu stručnjaci, mjeseci ustrajnog rada dok se u oku nisu stvorile dostatne količine rodopsina. Prvi znak da će terapija uspjeti bio je kada je pacijent prilikom šetnje odjednom počeo raspoznavati oblike na pješačkom prijelazu. 

Dr. José-Alain Sahel s pariškog Instituta za vid rekao je kako je pacijent isprva bio pomalo frustriran zbog činjenice da postupak traje toliko dugo. 

- Ali, kao što možete zamisliti, bio je vrlo uzbuđen kad je spontano počeo raspoznavati svari. Svi smo bili uzbuđeni. Još uvijek nema sjajan vid, no razlika između nepostojećeg i ograničenog vida nešto je što mijenja život - kazao je liječnik. 

Goleme mogućnosti

Botond Roska, sa sveučilišta u švicarskom Baselu rekao je kako nova saznanja dokazuju da je optogenetika terapija budućnosti kada je povrat vida u pitanju. 

Ipak, to je samo jedna od brojnih tehnika koje liječnici diljem svijeta isprobavaju kako bi osobama vratili vid, piše BBC. Mnoge se također bave popravkom genetskih problema koje uzrokuju probleme s vidom, no s retinitis pigmentosom to je posebno teško jer ju može uzrokovati mutacija više od 71 različitog gena. Neki znanstvenici pokušavaju oboljelima vratiti vid tako što povezuju videokameru s elektrodama implantiranim u pozadinu oka. 

Vjeruje se da su mogućnosti primjene optogenetike goleme, a neki zagovaraju upotrebu tehnike pri liječenju Parkinsonove bolesti, pa čak i kod nadvladavanja problema uzrokovanih moždanim udarom. 

Britanski profesor i stručnjak za bolesti mrežnice James Bainbridge, ustvrdio je kako je studija izuzetno obećavajuća, no da tek treba vidjeti rezultate na drugim ispitanicima, jer je sada provedena samo na jednom pacijentu.

Francuz već više od 40 godina boluje od genetski uvjetovanog očnog stanja, retinitis pigmentosa, a koje uzrokuje umiranje fotoosjetljivih stanica, piše Jutarnji list.

Znanstvenici su došli do otkrića koje bi moglo olakšati život milijuna ljudi diljem svijeta - potpuno slijepom muškarcu vid je djelomično vraćen nakon što je podvrgnut terapiji proteinom koji je prvotno otkriven kod algi. Biološka terapija, takozvana optogenetika, poseban je proces pri kojem se proteini koriste kako bi se organizmu vratila kontrola nad stanicama u pozadini oka.
