Tvar otkrivena u šipku može ojačati imunološke stanice koje pomažu u borbi protiv raka, pokazalo je novo istraživanje provedeno na Sveučilištu u Frankfurtu.

Istražujući terapije za rak debelog crijeva, znanstvenici su otkrili da metabolit u šipku, poznat kao urolitin A, pomlađuje imunološke T stanice koje su tada učinkovitije u borbi protiv tumora.

Rak debelog crijeva i rektuma i dalje je bolest s visokom stopom mortaliteta u kasnijim stadijima.

Rano otkrivanje i početak liječenja posljednjih su godina unaprijeđeni, no ne reagiraju svi pacijenti jednako dobro na nove terapije.

Trenutna istraživanja sugeriraju da je jedna od karakteristika tumora kao bolesti imunološka disfunkcija. Imunološke stanice koje bi se trebale boriti s tumorom potisnute su i blokirane okolnim tumorskim tkivom, njegovom mikrookolinom, piše Živim.

Posljedica toga je da su T stanice, koje su inače prirodan imunološki odgovor tijela na rak, ograničene pa je tumoru omogućeno da nekontrolirano raste i širi se. Tim znanstvenika pod vodstvom profesora Floriana Gretena u novoj je studiji došao korak bliže rješenju tog problema.

U časopisu Immunity objavili su studiju koja je pokazala da urolitin A reciklira i obnavlja mitohondrije, takozvane „elektrane” unutar T stanica – kroz proces poznat kao mitofagija. Uvođenjem ekstrakta šipka uklanjaju se stari i oštećeni mitohondriji u T stanicama i zamjenjuju novim i funkcionalnim. To mijenja genetski sastav T stanica koje su tada sposobnije za uništavanje tumora. Autor studije dr. Dominic Denk iz Sveučilišne bolnice u Frankfurtu kaže da je njihovo otkriće vrlo uzbudljivo jer fokus nije na tumorskoj stanici već na imunološkom sustavu, prirodnoj obrani od raka.

- Još uvijek nedostaju pouzdani terapijski pristupi u stvarnosti pacijenata s kolorektalnim tumorom. Poboljšanjem kombinirane terapije s postojećim imunoterapijama, ova studija otvara značajne nove mogućnosti kliničke primjene.

Nadamo se da ćemo ovo iskoristiti za održivo poboljšanje u liječenju raka debelog crijeva, ali i drugih vrsta tumora - izjavio je dr. Denk.

Terapijski potencijal 

Terapijski potencijal urolitina A znanstvenici su prikazali na dva načina. Urolitin A se može unijeti u organizam koristeći šipak kao hranu u pretkliničkom modelu, što ograničava rast tumora i čak djeluje sinergijski s postojećom imunoterapijom.

S druge strane, dobrobiti urolitina A uočene su i na ljudskim T stanicama u laboratoriju. In vitro tretman urolitinom A pomlađuje ljudske T stanice tako što proizvodi T memorijske matične stanice – moćne imunološke matične stanice koje zbog svoje sposobnosti diobe stalno opskrbljuju imunološki sustav pomlađenim T stanicama.

Na temelju ovih otkrića znanstvenici planiraju primijeniti urolitin A u kliničkim ispitivanjima terapija za osobe s rakom debelog crijeva. Prof Greten, koji je ujedno i glasnogovornik Instituta za rak u Frankfurtu, kaže da je ova studija još jednom pokazala kako su uspješni interdisciplinarni pristupi frankfurtskog Instituta za rak.

- Izuzetno smo zadovoljni što ćemo naše rezultate vrlo brzo pretočiti u kliničku primjenu i s uzbuđenjem očekujemo klinička ispitivanja koja su pred nama - zaključio je.

Moćan je

Šipak je bogat izvor flavonoida, selena, magnezija te vitamina iz grupe E, A i B kompleksa. Pomaže u snižavanju krvnog tlaka, kolesterola, potiče rad prostate, ali i povećava plodnost kod žena.

Tvar otkrivena u šipku može ojačati imunološke stanice koje pomažu u borbi protiv raka, pokazalo je novo istraživanje provedeno na Sveučilištu u Frankfurtu.

Istražujući terapije za rak debelog crijeva, znanstvenici su otkrili da metabolit u šipku, poznat kao urolitin A, pomlađuje imunološke T stanice koje su tada učinkovitije u borbi protiv tumora.

Rak debelog crijeva i rektuma i dalje je bolest s visokom stopom mortaliteta u kasnijim stadijima.

Rano otkrivanje i početak liječenja posljednjih su godina unaprijeđeni, no ne reagiraju svi pacijenti jednako dobro na nove terapije.

Trenutna istraživanja sugeriraju da je jedna od karakteristika tumora kao bolesti imunološka disfunkcija. Imunološke stanice koje bi se trebale boriti s tumorom potisnute su i blokirane okolnim tumorskim tkivom, njegovom mikrookolinom, piše Živim.

Posljedica toga je da su T stanice, koje su inače prirodan imunološki odgovor tijela na rak, ograničene pa je tumoru omogućeno da nekontrolirano raste i širi se. Tim znanstvenika pod vodstvom profesora Floriana Gretena u novoj je studiji došao korak bliže rješenju tog problema.

U časopisu Immunity objavili su studiju koja je pokazala da urolitin A reciklira i obnavlja mitohondrije, takozvane „elektrane” unutar T stanica – kroz proces poznat kao mitofagija. Uvođenjem ekstrakta šipka uklanjaju se stari i oštećeni mitohondriji u T stanicama i zamjenjuju novim i funkcionalnim. To mijenja genetski sastav T stanica koje su tada sposobnije za uništavanje tumora. Autor studije dr. Dominic Denk iz Sveučilišne bolnice u Frankfurtu kaže da je njihovo otkriće vrlo uzbudljivo jer fokus nije na tumorskoj stanici već na imunološkom sustavu, prirodnoj obrani od raka.

- Još uvijek nedostaju pouzdani terapijski pristupi u stvarnosti pacijenata s kolorektalnim tumorom. Poboljšanjem kombinirane terapije s postojećim imunoterapijama, ova studija otvara značajne nove mogućnosti kliničke primjene.

Nadamo se da ćemo ovo iskoristiti za održivo poboljšanje u liječenju raka debelog crijeva, ali i drugih vrsta tumora - izjavio je dr. Denk.

Terapijski potencijal 

Terapijski potencijal urolitina A znanstvenici su prikazali na dva načina. Urolitin A se može unijeti u organizam koristeći šipak kao hranu u pretkliničkom modelu, što ograničava rast tumora i čak djeluje sinergijski s postojećom imunoterapijom.

S druge strane, dobrobiti urolitina A uočene su i na ljudskim T stanicama u laboratoriju. In vitro tretman urolitinom A pomlađuje ljudske T stanice tako što proizvodi T memorijske matične stanice – moćne imunološke matične stanice koje zbog svoje sposobnosti diobe stalno opskrbljuju imunološki sustav pomlađenim T stanicama.

Na temelju ovih otkrića znanstvenici planiraju primijeniti urolitin A u kliničkim ispitivanjima terapija za osobe s rakom debelog crijeva. Prof Greten, koji je ujedno i glasnogovornik Instituta za rak u Frankfurtu, kaže da je ova studija još jednom pokazala kako su uspješni interdisciplinarni pristupi frankfurtskog Instituta za rak.

- Izuzetno smo zadovoljni što ćemo naše rezultate vrlo brzo pretočiti u kliničku primjenu i s uzbuđenjem očekujemo klinička ispitivanja koja su pred nama - zaključio je.

Moćan je

Šipak je bogat izvor flavonoida, selena, magnezija te vitamina iz grupe E, A i B kompleksa. Pomaže u snižavanju krvnog tlaka, kolesterola, potiče rad prostate, ali i povećava plodnost kod žena.
